132 JUDIT NAGY et al. tion controls. The specificity of immune sera was checked by immune diffusion. Immunofluorescence revealed large amounts of granular type IgG (Fig. 2) and HBS antigen (Fig. 3) along the basement membrane of all glomeruli. IgM and C3 could also be observed in a similar pattern although in lower amounts along the glomerular basement membrane (Fig. 4). In the sections incubated with IgA, the basement membrane of all glomeruli showed intensive linear staining. At higher magnification this was made up of confluent, tightly packed granules (Fig. 5). Electron microscopy. Specimens 1—2 mm in diameter were cut from the kidneys fixed in formalin. Tw enty-four hours washing in buffered solution was followed by fixation in osmium tetroxide buffered with 1% phosphate buffer pH 7.4 at 4°C for 90 min. The material after dehydration was embedded in Durcupan. Semi-thin and ultra-thin sections were cut with Ultrotome III LKB ultramicrotome. Semi-thin sections were stained with haematoxylin—eosin periodic acid methenamine silver and PAS. Ultrathin sections were contrasted with uranyl acetate and lead citrate. Electron microscopic examinations were done with TESLA BS 242 E electron microscope. The semi-thin sections, particularly when stained with periodic acid— methenamine-silver, demonstrated well the spikes on the epithelial side of the glomerular basement membrane that gave a peculiar thorny appearance to the membrane (Fig. 6).
